Cell and Gene Therapy

The Moxi Go II has been valuable for our day-to-day experimentation and testing. We use the system for bi-weekly QC to test the efficiency of binding of our targeting-fusion protein in each batch we create. This provides a fast and cheap way to optimize and maintain quality control for down stream applications in translational discovery

Traditional Cell Analyzer are not practical in this environment given their large footprint, heavy maintenance and cleaning burden, production of significant fluid waste and difficulty of operation. Additionally traditional Cell Analyzer do not integrate the Coulter Principle, which provides a unique window into immune cells and their cell volume. ORFLO has addressed many of these shortcomings​

In the CAR-T world monitoring the cell volume during T cell expansion is essential to assess the activation state of our CAR-T, i.e., the bigger, the more activated. We have defined thresholds to define when a T cell is resting down. Having cell concentration, volume and viability in a single count is ideal for our work.​

Moxi Go II enables me to rapidly count viable primary murine splenocytes while also enabling quantification of the CD4+ (or CD8+) T cell compartment — key information I need to set up complex co-culture assays. Since all of the cells I work with are from primary sources, speed is critical when setting up experiments to ensure maximum viability. The Moxi Go II significantly reduces my reliance on traditional Cell Analyzer for experiment set up and QC.​

Having a Target to effector ratio that is off from one day to the next can lead to a lot of variability in results. The Moxi minimizes this variability. In these types of experiments, it is nice to not have to worry about T:E ratios and have it controlled experiment to experiment. It is also useful to have accurate counts when you are doing any type of experiments using Moxi’s impedance-based readout.​
